I am in the process of migrating from Exchange 2010 to Exchange 2016 in a single DAG two server environment.

I want to use CommVault to perform mailbox level backups and also need to truncate the Exchange 2016 database logs.

Would I need to configure both database and mailbox agents, or can the mailbox agent truncate the DB logs?

Exchange Mailbox backup does not truncate the DB Logs.  You will need to use the DB Agent for that.
